Ple151-EGFP Description:

The Ple151 promoter construct is derived from the promoter region of the oligodendrocyte transcription factor 1 (OLIG1) gene. The Pleiades Promoter Project created a 3033 bp MiniPromoter derived from one upstream and two downstream putative regulatory subregions along with a section of the promoter, and introduced them into the multiple cloning site (MCS) of Pleiades expression vector pEMS1306 (EGFP reporter flanked by minimal frt sites) to make plasmid pEMS1172 with Ple151 driving EGFP. The Ple151-EGFP (pEMS1172) was introduced by homologous recombination as a single copy insertion into the Hprt1b-m3 locus in the X chromosome of mouse embryonic stem cells (ESCs) mEMS1204.03A. Four ESC lines (mEMS1608, mEMS1609, mEMS1614, and mEMS1615) were PCR validated. Chimeras were generated using mEMS1615.

Note: Promoterless negative controls (pEMS1302, pEMS1306, pEMS1313) have been generated, as well as CAG positive controls (pEMS1157, pEMS1277 & pEMS1488).

 

Ple151-EGFP Expression Pattern:

Positive EGFP immunoreactivity was detected across numerous brain regions. EGFP-positive cells show darker staining in the nucleus with a lighter staining halo of processes. In the brainstem, cortex and thalamus, numerous immunopositive profiles are detected while fewer profiles are seen in the olfactory bulb, striatum and cerebellum. In contrast to the predicted expression of Olig1, the EGFP expression does not appear to be enriched in white matter tracts such as the corpus callosum.
