Ple88 Description:

The Ple88 promoter construct is derived from the promoter region of the Glial Fibrillary Acidic Protein gene (GFAP). The Pleiades Promoter Project isolated a 2210 bp subregion of the GFAP promoter immediately upstream of the first exon of the gene and introduced it into the multiple cloning site (MCS) of Pleiades expression vector pEMS1306 (EGFP reporter flanked by minimal frt sites) to make plasmid pEMS1375 with Ple88 driving EGFP. The Ple88-EGFP (pEMS1375) was introduced by homologous recombination as single copy insertion into the Hprt1b-m3 locus in the X Chromosome of mouse embryonic stem cells (ESCs) mEMS1204. The resulting PCR validated four ESC lines, mEMS1653, mEMS1661, mEMS1671, and mEMS1672. Chimeras were generated using mEMS1653 and this cell line was used to generate the images above.

Note: a promoterless negative control (pEMS1306) has been generated, as well as a two CAG positive controls (pEMS1157 & pEMS1277).
