Ple34-lacZ Description:

This Ple34 promoter construct is derived from the promoter region of the Claudin 5 (CLDN5) gene. The Pleiades Promoter Project created a 3845 bp MiniPromoter derived from two upstream putative regulatory subregions along with a section of the promoter and introduced it into the multiple cloning site (MCS) of Pleiades expression vector pEMS1313 (LacZ reporter flanked by full frt sites (WT and F5 mutant)) to make plasmid pEMS1505 with Ple34 driving lacZ. The Ple34-lacZ (pEMS1505) was introduced by homologous recombination as single copy insertion into the Hprt1b-m3 locus in the X Chromosome of mouse embryonic stem cells (ESCs) mEMS1202. The resulting PCR validated four ESC lines, mEMS4613, mEMS4617, mEMS4598, and mEMS4609. Chimeras were generated using mEMS4613.

Note: Promoterless negative controls (pEMS1302, pEMS1306, pEMS1313) have been generated, as well as CAG positive controls (pEMS1157, pEMS1277 & pEMS1488).
