Ple55-lacZ Description:

The Ple55 promoter construct is derived from the promoter region of the Doublecortin (DCX) gene. The Pleiades Promoter Project created a 3877 bp MiniPromoter derived from one downstream putative regulatory subregion along with a section of the promoter and introduced them into the multiple cloning site (MCS) of Pleiades expression vector pEMS1313 (LacZ reporter flanked by full frt sites (WT and F5 mutant)) to make plasmid pEMS1526 with Ple55 driving lacZ. The Ple55-lacZ (pEMS1526) was introduced by homologous recombination as single copy insertion into the Hprt1b-m3 locus in the X Chromosome of mouse embryonic stem cells (ESCs) mEMS1202. The resulting PCR validated four ESC lines, mEMS3879, mEMS3870, mEMS3860, and mEMS3884. Chimeras were generated using mEMS3879.

Note: Promoterless negative controls (pEMS1302, pEMS1306, pEMS1313) have been generated, as well as CAG positive controls (pEMS1157, pEMS1277 & pEMS1488).
